Doxorubicin metabolism pathway
WebSep 10, 2024 · Doxorubicin exerts anti-cancer biological activity by mechanisms involving intercalation into DNA and impairing topoisomerase-II [ 1, 2, 3 ]. Despite its wide implementation in various cancer therapy regimens, its dosage is limited by cardiotoxicity and other organ toxicity [ 4, 5, 6, 7 ]. WebDoxorubicin (DXR) is an anthracycline-type polyketide, typically produced by Streptomyces peucetius ATCC 27952. Like the biosynthesis of other secondary metabolites in Streptomyces species, DXR biosynthesis is tightly regulated, and a very low level of DXR production is maintained in the wild-type strain.

WebThe CYP450 induction metabolism assays indicate that AHCC is an inducer of CYP450 2D6. AHCC does have the potential for drug-drug interactions involving CYP450 2D6, such as doxorubicin or ondansetron; however, the overall data suggest that AHCC would be safe to administer with most other chemotherapy agents that are not metabolized via the ... WebJul 1, 2024 · Importantly, genetic mutation of cyp1a protected zebrafish against doxorubicin-induced cardiotoxicity phenotypes. Together, these results provide strong evidence that CYP1 is an important contributor to doxorubicin-induced cardiotoxicity and highlight the CYP1 pathway as a candidate therapeutic target for clinical cardioprotection.
